What This Skill Does
Query clinical variant data from ClinVar and gnomAD via Clarity Protocol. Returns pathogenicity classifications, review status, allele frequencies, and submission metadata for variants in a given gene.
Combines ClinVar classification and gnomAD frequency data in a single query instead of checking two databases separately.
When to use it
- Check if a BRCA2 variant is classified as pathogenic in ClinVar
- Look up gnomAD population frequency for a candidate disease variant
- Review ClinVar submission quality before reporting a variant
- Filter rare vs common variants across a gene's clinical record
- Compare clinical significance across multiple variants in a gene
Example Workflow
Here's how your AI assistant might use this skill in practice.
User asks: What is the clinical significance and population frequency of NM_005910.6:c.926C>T in MAPT?
- 1Query the Clarity Protocol API with the specified gene symbol and variant notation
- 2Retrieve ClinVar significance, review status, and last evaluation date
- 3Retrieve gnomAD allele frequency and allele count
- 4Interpret frequency against gnomAD thresholds (very rare, rare, uncommon, common)
- 5Format results into a readable summary
Variant NM_005910.6:c.926C>T in MAPT is classified as Pathogenic with criteria provided by multiple submitters; gnomAD AF is 0.000004 (very rare, < 0.01%)
